Stratagene's "Quik Solution"

The "Quik Solution" is a component of Stratagene's various site-specific mutagenesis kits.

 

It is certainly a pure DMSO!

 

Evidence:

- Smells like DMSO;

- Freezes at +18C, just like DMSO;

- DMSO was tested to be equivalent to the Quik Solution in mutagenesis reactions;

- DMSO is a frequent additive to various termostable polymerase-based applications.

 

Note: In general, the kits don't contain anything that you can't obtain much cheaper separately. Just buy Stratagene's Pfu Ultra polymerase, make your electroporation competent cells and, provided that you have dNTPs, DMSO, Dpn I from NEB and primers, you have everything to perform the reactions according to the manual.

Hints: reduce volume to 25 ul and use 0.5 ul polymerase per reaction; for Dpn I treatment it is better to add 1 ul of Dpn I to a 10 ul reaction (instead of full reaction volume).
